Gene-disease: first associated with skeletal dysplasia in 1993. Phenotypes: pseudoachondroplasia and multiple epiphyseal dysplasia with no clear genotype-genotype correlation. At least 15 unique variants (missense, inframe indels) in 15 individuals in 10 papers. Segregated with diease. Supported by mouse models, expression studies and functional studies. Green on Skeletal dysplasia panel. Definitive ClinGen association with skeletal dysplasia. Fetal phenotype: PMID 39521787 fetal case (Fetal short long bones (<-5SD), bowing of the legs, genu varum, COMP missense variant inherited from affected father). PMID 40188065: 1 fetus with maternally inherited *frameshift* variant and fetal growth restriction, maternal phenotype not stated.
