Stephanie Allen (Consultant Clinical Scientist)

Green List (high evidence)

This gene and phenotype were reviewed during a meeting on 2nd March 2023 between representatives of the North Thames and Central & South R21 testing GLHs. Clinical review and curation was performed by Lyn Chitty, (North Thames GLH), and Stephanie Allen, Denise Williams, Anna de Burca and Megan Horton-Bell (Central & South GLH). Outcome of review: Confirmed that the phenotype is fetally relevant, support adding to the Fetal anomalies panel as a Green gene.

On 7 panels, inc. IEM, severe paediatric disorders, DDG2P. Associated with Mitochondrial complex I deficiency, nuclear type 33 (AR). Alston et al., 2018 PMID 30245030: Subject 1: prenatal scan: IUGR, intracranial ventriculomegaly. Fetal MRI at 30wks confirmed ventriculomegaly with mild cerebellar hypoplasia. c.191G>C p.(Arg64Pro) and c.265G>T p.(Glu89*) compound het. Subject 3: prenatal: oligohydramnios and IUGR. At birth: dysmorphic features. c.3G>A p.(?) hom. Conclusion: link to prenatal phenotypes (IUGR, oligohydramnios, ventriculomegaly). Relevant
