20 patients from 18 families with rare missense variants clustered in specific protein regions (confirmed de novo in 12) and phenotypes inluding intrauterine growth retardation, dysmorphism, bone dysostosis, neuroanatomical abnormalities (dysgenesis of the corpus callosum, pituitary hypoplasia with growth hormone deficiency, Chiari I malformation) and variable birth defects including cardiac defects, talipes. Postnatally: short stature, neurodevelopmental delay, immunodeficiency.
