Candidate gene contributing to DiGeorge syndrome phenotype (aka TUPLE1). De novo heterozygous truncating variants associated with a neurodevelopmental disorder in two unrelated patients in 33417013. De novo start loss variant in a newborn with tetralogy of Fallot, corpus callosum hypoplasia, and phenotypic features similar to DiGeorge syndrome in 38511226. LoF variants found at very low frequency in UK Biobank cohort suggesting incomplete penetrance (34074949). Amber on ID panel.
