This gene and phenotype were reviewed during meetings between November 2024 & January 2025. Missense variants within actin-binding domains with evidence of gain-of-function effect reported in 8 unrelated families with an X-linked condition characterized by diaphragm defects, variable anterior body-wall anomalies, and/or facial dysmorphism (37751738). Knock-in mouse model recapitulates phenotype (37751738). One further family reported - missense variant detected in fetus with CDH and other anomalies and in previous pregnancy losses (39432562). Loss-of-function variants are associated with childhood-onset X-linked osteoporosis; no reports of antenatal presentation. Only gain-of-function variants of clear relevance prenatally.
