This gene and phenotype were reviewed during meetings between November 2023 & July 2024. The meetings included representatives of the North Thames and Central & South R21 testing GLHs and from the R21 Clinical Oversight Group. Outcome of review: Confirmed that the phenotype is fetally relevant, support adding to the Fetal anomalies panel as a Green gene.

SPEN is on the paediatric disorders super panel. It is also green on the intellectual disability panel. This is an AD condition. PMID:33596411- This paper outlines the phenotype of 34 patients with Loss of Function variants on SPEN. Of these 64% (14/22) had CNS anomalies-, 64% had congenital heart disease (14/22/) and 21% had kyphosis. De novo truncating variants present with Multiple congenital anomalies, dysmorphism, DD, ID. Green given MCA whihc could be identified on prenatal untrasound/MRI. Green
