Homozygous missense variants p.Ser272Thr and p.Ala2Val in 22 infants from 3 consanguineous families with cerebellar and corpus callosum hypoplasia, abnormal myelination of the central nervous system or spinal motor neuron disease (24989451). Homozygous missense variant p.Val80Ile causing exon skipping in a patient with psychomotor retardation, spasticity, spinal muscle atrophy, respiratory problems, diaphragmatic hernia, dilated lateral ventricles, hypoplastic temporal lobes, thinning of the brain stem, dysmorphic facies, nystagmus, congenital esotropia and contractures (38017281). Milder phenotype in a patient with compound heterozygous splice and p.Ser272Thr variants (34210538). Now rated green on DDG2P.
