Recent publication (2021) identified by Zornitza Stark (https://doi.org/10.1016/j.xhgg.2020.100015) reports 7 unrelated individuals with missense variants in this gene. All patients presented with DD/ID and hypotonia, and all those of school age have attended special school and have significant ID.

Removed 'for-review' tag as this new evidence supports the current Green gene rating on this panel and demonstrates the role of intragenic variants in this disorder.

There are many cases of 2q37.3 terminal region (includes HDAC4) loss in PMID 30848064, however, there are only two intragenic variants in HDAC4, with a rating of VUS.
This gene should be rated as amber at the next major review.
